Unless you open the course in a chromeless window (wouldn't recommend it - doesn't work in XP / IE SP1 or better) you are not going to be able to remove the close button. Generally a bad idea security wise.

Best bet is to put good resume functions in for those that close out inadvertently. I've also gotten to the point where I have ZERO UI elements near the top of the screen. This keeps the mouse out of the top 10% of the screen. Also avoid popping up a heap of elements near the same size as the course presentation window and avoid using instructions like 'Click exit' or other directives that a user could easily confuse with the close button.
